buka n pasang bioreactor tu dengn bantuan Mr. shaman..
thank 4 the treat..
hari 1st cuma pasang komponen2 tok bioreactor tu
bioreactor yg siap dipasang..
n sediakan media tok yeast
preparation media for yeast we used YEPG medium
2% yeast extract
2% peptone
1% glucose
for 200ml
ingredients used
to incubate overnight 37 degree 200rpm
Day 2, 5 Oct 2010, 830am-5pm
inoculate 1 loopful colony of S.Cerevisiae
under aseptic technique
inoculate into media
the yeast was incubate overnight 30 degree 200rpm
preparation media for the bioreactor..
1500ml
used same recipe as the yeast media
before
after
pour the media into the bioreactor
autoclave..15min 121 degree
why 15 min?not above 15 min
to reduce the caramelize
Day 3, 6 Oct 2010, 10-5pm
taking the 0th of the sample..
as it before pour the yeast into the fermenter
the taking smple after pour the inoculum.
then take sample every each 1 hour..
for night...after 12 take smple every each 2 hour

Day 4, 7Oct 2010, 11-5pm
today
is the last day..
disposal techniques..
because we only use S.cerevisiae
n it a wild type n not harmful
so just pour it into the sink...
and wash all the equipment of the bioreactor...
last week...
is really the most busy and tired...
uwaaa...
even penat pn..thank to Mr. Shaman for the treat at the KFC
